成纤维细胞生长因子21(FGF21)与FGF19和FGF23一起是FGF超家族的内分泌成员。已被证明可以作为代谢调节剂,在控制葡萄糖稳态,胰岛素敏感性和生酮作用方面发挥作用。肝脏被认为是FGF21产生的主要部位,并释放到血液中。此外,肝外组织,如白色和棕色脂肪组织和骨骼肌,也表达FGF21。


FGF21对靶细胞的作用需要FGFRs和β-Klotho,这是一种单通道跨膜蛋白,可作为FGF21信号传导的受体因子。当两者结合时,FGF21会刺激胰岛素敏感性和葡萄糖代谢,从而导致体重下降。通过FGF21给药处理或者体内过表达FGF21,能够降低肥胖的啮齿类动物、猴类以及人类罹患高胰岛素血症、胰岛素抵抗和高血糖等疾病风险。
