胰高血糖素样肽-1受体(GLP-1R)是在胰腺细胞和大脑神经元上发现的一种受体蛋白。 在人类中,它是由位于6号染色体上的GLP1R基因合成的。 它是G蛋白偶联受体中胰高血糖素受体家族的成员。


被激活的GLP-1R刺激腺苷酸环化酶通路,导致胰岛素合成和胰岛素释放增加。因此,GLP-1R一直是开发治疗糖尿病药物(通常称为GLP1R激动剂)的目标。GLP-1R也在大脑中表达,参与控制食欲。


GLP-1R是一种多效性偶联受体, N端识别特异性的配体,通过与多种G蛋白(Gαs、Gαi、Gαo和Gαq/11)偶联来调控细胞通路。当与GLP-1结合后,G蛋白α亚基与β、γ亚基解离,Gαs蛋白偶联,激活腺苷环化酶(cAMP),促使cAMP在细胞内含量升高并增加蛋白激酶A(PKA)的含量,激活下游信号通路,引起胰岛素基因转录增加。


吉满生物H_GLP1R Reporter CHO-K1 Cell Line报告基因细胞系,是基于cAMP-PKA信号通路构建的一种Luciferase报告基因细胞系。当GLP-1与其受体结合时,亚基互相解离。Gαs蛋白偶联,激活腺苷环化酶,促使cAMP在细胞内含量升高并增加PKA的含量。激活下游信号通路,从而激活荧光素酶(Luciferase)的表达。Luciferase读值即代表信号通路的激活效果,因此可用于GLP1R相关药物的体外效果评价。
