产品简介

IFN-α由多种细胞类型分泌,通过刺激巨噬细胞和 NK 细胞引发抗病毒反应,且具有抗肿瘤活性。IFN-α通过改变下丘脑中热敏神经元的活动而充当致热因子,从而引起发烧。运用类似的机制,IFN-α可以用来减轻疼痛,与 μ-阿片受体相互作用,起到镇痛剂的作用。


所有 I 型干扰素 (IFN:IFN-α、IFN-β、IFN-ε、IFN-κ和IFN-ω)都与人体细胞表面的一个共同受体结合,该受体被称为 I 型 IFN 受体。I 型 IFN 受体由两个亚基 IFNAR1 和 IFNAR2 组成,它们分别与 Janus 活化激酶 (JAK) 酪氨酸激酶 2 (TYK2) 和 JAK1 相关。与 I 型 IFN 受体相关的 JAK 的激活导致 STAT2和 STAT1 的酪氨酸磷酸化;这导致 STAT1-STAT2-IRF9复合物的形成,它们被称为 ISGF3(干扰素刺激基因 (ISG) 因子 3)复合物。这些复合物易位到细胞核并结合 DNA 中的 IFN 刺激反应元件以启动基因转录。


吉满生物IFNα Reporter THP1 Cell Line报告基因细胞系,是基于STATs信号通路构建的一种Luciferase报告基因细胞系。当IFN-α结合I 型 IFN 受体( IFNAR1 和 IFNAR2 )后,导致信号通路激活,形成ISGF3复合物,复合物易位到细胞核并结合DNA中的IFN 刺激反应元件,从而激活荧光素酶(Luciferase)的表达。Luciferase读值即代表信号通路的激活效果,因此可用于IFNα相关药物的体外效果评价。
