PD-1是激活的T细胞和B细胞表达的一种免疫抑制性受体,在对肿瘤抗体和自身抗原的免疫反应的调节中起关键作用。邻近细胞间的PD-1与其配体PD-L1或PD-L2的相互作用会抑制TCR信号通路的传导以及TCR介导的细胞增殖、转录激活和细胞因子产生等效应。用于阻断PD-1/PD-L1相互作用的治疗抗体和Fc融合蛋白在治疗各种癌症的临床试验中已表达出很好的应用前景。目前用于检测anti-PD-1或anti-PD-L1生物制品活性的方法依赖于初级人类T细胞和功能终点的测量,如细胞增殖、细胞表面标志物表达、干扰素γ(IFNγ)和白介素-2 (IL-2)的产生。由于依赖于供体原代细胞、复杂的试验方案和不合格的试验试剂,这些试验既费力又易变。因此,这些测定方法很难在质量控制的药物开发环境中建立。


吉满生物Mouse_PD-1 Reporter Jurkat Cell Line报告基因细胞系,该细胞稳定表达Mouse_PD-1基因及Luciferase报告基因。其配套细胞Mouse PDL1 aAPC CHO-K1 Cell Line (Genomeditech/GM-C25791),是一种稳定表达小鼠PD-L1和一种以抗原非依赖方式激活同源TCR的细胞表面蛋白的CHO-K1细胞。当两种细胞共培养时,PD-1/PD-L1之间的相互作用会抑制TCR信号通路的转导及转录因子介导的luciferase表达。加入阻断PD-1/PD-L1的抗体后,这种抑制会被解除,引起TCR信号通路的传导及转录因子介导的luciferase的表达,可用于测定阻断PD-1/PD-L1相互作用的抗体及其他生物制剂的效能和稳定性。
